dc.description.abstract Polyglutamine (polyQ) diseases are caused by expansion of glutamine (Q) repeats within the associated protein that induces formation of the intracellular protein aggregates. Collected evidence to date suggests that strong tendency of these mutated proteins toward forming oligo-mers and/or aggregates together with their numerous targets may be the primary cause of the tox-icity causing neuropathy. However, it remains largely unclear what characteristics of the pathogenic polyQ proteins cause these inclusion bodies and how these properties contribute to the various aspects of the disease pathogenesis. Here, we studied what properties of the pathogenic polyQ proteins are primarily associated with the specific early neuropathy, the distal dendrite defects. Different types of structural variants of polyQ proteins had been generated, and only structural variants possessing coiled coil propensity were able to induce the dendrite defects, accompanied by their preferred localization in the nucleus. Given that total mRNA level was decreased by expression of the pathogenic SCA3 accumulated within the nucleus, the observed early neuropathy appeared to be closely associated with the changes in the nuclear transcriptome. Furthermore, we demonstrated that Q continuity within the associated protein is also required for the dendrite abnormalities. In addition, coiled coil property of toxic SCA3 proteins seemed to be critical for interactions with their potential targets as well as their self-aggregation. Taken together, both coiled coil property and Q continuity may be the key contributors necessary for inducing distal dendrite defects caused by pathogenic SCA3. ⓒ 2015 DGIST -

dc.description.alternativeAbstract 폴리글루타민 질환 (polyglutamine disease)의 발병은 해당 질환과 관련된 단백질의 아미노산 서열에서 글루타민의 수가 증가되는 것에서 기인하며, 이 질환 단백질은 세포 내 단백질 응집체 (protein aggregates) 형성을 주도하는 것으로 알려져 있다. 비록 지금까지 논쟁이 계속되고 있지만, 단백질 응집체의 형성 과정에서 표적 단백질 (target proteins)을 포획한 폴리글루타민 단백질 응집체가 세포 내 독성 (intracellular toxicity) 작용과 밀접한 관련성이 있다는 것이 다수의 기존 연구에서 보고되어 왔다. 그러나 여전히 병원성 폴리글루타민 단백질의 어떠한 구조적 특성이 폴리글루타민 단백질 응집체 형성의 주요한 원인이 되는지, 그리고 이 단백질의 구조적 특성이 폴리글루타민 질환의 증상에 어떠한 영향을 주는지는 분명히 밝혀진 바가 없다. 따라서 본 연구에서는 폴리글루타민 질환의 초기 증상 중 말단 수상 돌기 손상 (dendrite defect)에 결정적인 영향을 끼치는 병원성 폴리글루타민 단백질의 구조적 특성을 밝혀내고자 하였다. 단백질의 2 차원 구조를 여러 조건으로 수정하여 만든 변형 단백질들 중, 알파-나선 구조의 감긴 코일 (coiled coil) 구조를 가지고 있는 단백질이 발현된 수상 돌기에서 형태적 변형이 관찰되었다. 그리고 매우 흥미롭게도, 수상 돌기 손상을 일으키는 감긴 코일형 폴리글루타민 단백질은 대부분 세포의 핵 내에서 발견되었다. 병원성 폴리글루타민 단백질 응집체가 핵 안에 위치하는 경우에만 핵 내에서 일어나는 유전자의 일반적인 전사 수준이 크게 감소하는 현상으로 판단해 볼 때, 관찰된 초기 신경 장애는 핵 내 전사체의 변화와 밀접한 관련이 있는 것으로 보여졌다. 이에 더하여 비정상적인 수상 돌기를 유발하는데, 폴리글루타민 단백질의 1 차 아미노산 서열에서 특정 개수 이상의 연속적 글루타민 (continuous glutamine) 존재가 전제되어야 한다는 것을 확인하였다. 또한 독성 폴리글루타민 단백질의 감긴 코일 구조는 해당 질환 단백질 자체의 응집체 형성 (self-aggregation) 뿐만 아니라, 이 단백질의 잠재적 표적 단백질과의 상호작용 (target-interaction)에도 영향을 주는 것으로 나타났다. 종합하면, 본 연구를 통해 병원성 폴리글루타민 단백질의 감긴 코일 구조와 연속적 글루타민 서열이 폴리글루타민 질환의 초기 증상인 말단 수상 돌기 손상을 유발하는데 결정적인 역할을 한다는 사실을 새롭게 밝혀내었다. ⓒ 2015 DGIST -
