| dc.description.abstract | 수정체는 높은 투명성과 굴절 기능을 유지해야 하는 특수한 조직으로, 수정체 섬유세포의 정교한 세포 구조와 장기간 안정적인 단백질 환경에 의해 그 기능이 유지된다. Filensin(BFSP1)과 phakinin(BFSP2)은 수정체 특이적 중간섬유 단백질로, 수정체 섬유세포에서 특화된 세포골격 구조를 형성하는 것으로 알려져 있다. 이들 단백질의 결손 또는 돌연변이는 수정체 섬유세포 구조 이상과 백내장 발생과 연관되어 있으나, filensin과 phakinin이 어떠한 분자적 원리에 따라 조립되고, 각 도메인이 필라멘트 형성과 안정성에 어떻게 기여하며, 백내장 관련 돌연변이가 이러한 조립 과정을 어떻게 방해하는지는 아직 충분히 규명되지 않았다. 본 연구에서는 filensin과 phakinin의 구조-기능적 관계를 규명하기 위해 세포생물학적 분석, 재조합 단백질 기반의 in vitro 재구성, 침강 분석, 투과전자현미경, CD 분광법, 유변학적 분석, pull-down assay, native PAGE, SEC–MALS 및 mass photometry를 이용하였다. 마우스 수정체의 면역조직화학 분석에서 filensin과 phakinin은 수정체 섬유세포층에서 중첩된 분포를 보였다. 또한 인간 수정체 상피세포주인 B-3 세포에서 filensin 또는 phakinin을 단독으로 발현시켰을 때는 확장된 필라멘트 구조가 형성되지 않았으나, 두 단백질을 함께 발현시켰을 때는 세포질 내 필라멘트성 구조가 형성되었다. 정제한 재조합 단백질을 이용한 in vitro 재구성에서도 filensin과 phakinin은 단독으로는 생산적인 필라멘트 구조를 형성하지 못했으며, 두 단백질을 함께 refolding했을 때 연속적이고 매끄러운 heteropolymeric filament를 형성하였다. Filament assembly의 stoichiometry를 분석한 결과, filensin과 phakinin은 equimolar stoichiometry에서 가장 효율적으로 sedimentable assembly에 incorporation되었으며, 이 조건에서 가장 길고 연속적인 필라멘트를 형성하였다. 반대로 stoichiometry가 equimolar 조건에서 벗어날 경우 필라멘트 길이가 감소하고 비필라멘트성 구조가 증가하였다. 특히 phakinin이 과량 존재하는 조건에서는 aggregate 형성이 두드러졌다. 이러한 결과는 filensin과 phakinin이 생산적인 필라멘트 형성을 위해 균형 잡힌 1:1 stoichiometry를 필요로 함을 시사한다. 수정체 단백질 환경과의 연관성을 조사하기 위해 αB-crystallin과의 관계를 분석하였다. αB-crystallin은 단독으로 또는 filensin 혹은 phakinin 단독 조건에서는 pellet fraction으로 잘 회수되지 않았으나, filensin과 phakinin이 함께 존재하여 필라멘트를 형성할 수 있는 조건에서는 pellet fraction에서 검출되었다. 또한 BSA는 filensin–phakinin assembly와 함께 sedimentation되지 않은 반면, lysozyme은 비특이적 aggregation과 관련된 pellet 형성을 유도하였다. 음성염색 TEM 분석에서 αB-crystallin이 없는 filensin–phakinin assembly는 smooth filament 형태를 보였으나, αB-crystallin을 첨가한 경우 beaded morphology가 관찰되었다. 이는 기존 문헌에서 보고된 수정체 “beaded filament” 형태가 filensin과 phakinin 자체의 고유한 형태라기보다, αB-crystallin과 같은 수정체 단백질의 결합 또는 decoration에 의해 나타날 수 있음을 시사한다. 도메인 절단 분석에서는 terminal domain이 filament assembly를 서로 다르게 조절한다는 것을 확인하였다. Head domain의 제거는 filament elongation을 감소시켰으며, 특히 phakinin head domain의 제거가 가장 큰 영향을 보였다. 반면 filensin tail domain의 제거는 filament 형성을 막지 않았고, in vitro 조건에서 filament morphology와 길이를 변화시켰다. 또한 young 및 aged mouse lens 모두에서 processed filensin species가 검출되었으며, aged lens에서 그 축적이 더 두드러졌다. 이는 filensin processing이 정상 렌즈에서도 존재할 수 있고, 노화에 따라 증가할 수 있음을 시사한다. Filensin과 phakinin의 core interaction module을 규명하기 위해 rod domain을 이용한 분석을 수행하였다. His-tagged filensin rod와 phakinin rod는 low-urea 또는 native-like 조건에서 결합하였으나, 높은 urea 농도에서는 안정적으로 결합하지 않았다. Ratio-dependent pull-down assay에서 phakinin rod가 과량 존재하더라도 elution fraction에서는 filensin rod와 phakinin rod가 약 1:1 비율로 회수되었으며, 남는 phakinin rod는 flow-through fraction에 남았다. 이는 두 rod domain이 refolding-dependent하고 saturable한 1:1 결합을 형성함을 시사한다. Native PAGE, chemical cross-linking 및 SEC–MALS 분석에서는 filensin–phakinin rod-domain complex가 약 318 kDa의 주요 species를 형성함을 보였으며, 이는 4:4 hetero-octameric assembly와 일치하는 결과이다. 따라서 filensin과 phakinin의 rod domain은 특이적 결합뿐 아니라 고차 heteromeric assembly 형성에 충분한 core module로 작용한다. 마지막으로 백내장 관련 돌연변이가 filensin–phakinin filament assembly에 미치는 영향을 분석하였다. Filensin D348N과 phakinin G112E, E233del, R287W, R339H를 포함한 변이체를 스크리닝한 결과, 대부분의 변이는 wild-type과 유사한 filament morphology, sedimentation profile 및 CD spectrum을 보였다. 그러나 phakinin E233del은 필라멘트 길이를 뚜렷하게 감소시켰으며, B-3 세포에서도 wild-type phakinin과 달리 filamentous network 대신 punctate structure를 주로 형성하였다. 또한 E233del-containing assembly는 shear stress 조건에서 WT assembly보다 더 빠르게 viscoelastic stability를 잃었고, temperature-dependent CD 분석에서도 낮은 apparent transition temperature를 보여 기계적 및 열적 안정성이 감소했음을 나타냈다. E233del의 결함이 glutamate side chain의 소실 때문인지, single-residue deletion 자체의 효과인지 구분하기 위해 E233A와 E233D 치환 변이체를 제작하였다. E233A와 E233D는 polypeptide chain length를 유지하면서 side-chain chemistry만 변화시키는 변이체이다. Full-length filament reconstitution에서 E233A와 E233D는 E233del 조건보다 긴 필라멘트 형성을 유도하였으며, 그 길이는 wild-type에 가까운 수준을 보였다. Rod-domain pull-down assay에서는 E233del이 filensin–phakinin의 1:1 결합을 소실시키지 않았으나, native PAGE와 mass photometry에서는 E233del 조건에서만 추가적인 high-molecular-weight heteromeric species가 관찰되었다. 이는 E233del이 초기 결합 자체를 방해하기보다는 higher-order oligomer heterogeneity를 증가시켜 productive filament elongation을 저해한다는 모델을 지지한다. 종합하면, 본 연구는 filensin과 phakinin이 수정체 특이적 중간섬유를 형성하기 위해 cooperative assembly, equimolar stoichiometry, rod-domain interaction, terminal-domain regulation을 필요로 함을 보여준다. 또한 αB-crystallin과의 association은 수정체 filament morphology에 영향을 줄 수 있으며, 노화에 따른 filensin processing은 수정체 중간섬유 시스템이 lens proteostasis와 밀접하게 연결되어 있음을 시사한다. 더불어 백내장 관련 phakinin E233del 변이는 1:1 결합을 직접적으로 파괴하기보다는 oligomeric assembly landscape를 변화시켜 필라멘트 elongation과 stress resistance를 저해하는 것으로 보인다. 본 연구는 수정체 특이적 중간섬유의 조립 원리와 백내장 관련 돌연변이의 분자적 영향을 이해하는 데 기초를 제공한다. 핵심어: 중간섬유, 수정체, filensin, phakinin, BFSP1, BFSP2, αB-crystallin, 백내장, filament assembly |The vertebrate ocular lens is a transparent and mechanically resilient tissue whose optical function depends on the highly ordered organization of lens fiber cells. Filensin and phakinin, encoded by BFSP1 and BFSP2, respectively, are lens-specific intermediate filament proteins that form a specialized cytoskeletal system in lens fiber cells. Although genetic and cellular studies have linked these proteins to lens transparency and cataract formation, the molecular principles governing their cooperative assembly, domain-level regulation, and mutation-associated defects remain incompletely understood. In this study, I investigated the structural and functional relationship of filensin and phakinin using a combination of cellular, biochemical, biophysical, and imaging-based approaches. Immunohistochemistry of mouse lens sections showed overlapping localization of filensin and phakinin in lens fiber cell layers. In human lens epithelial B-3 cells, neither filensin nor phakinin alone formed extended filamentous networks, whereas co-expression of the two proteins produced cytoplasmic filament structures. Consistently, in vitro reconstitution using purified recombinant proteins showed that filensin and phakinin cooperatively assemble into smooth heteropolymeric filaments, while each protein alone failed to produce productive filamentous structures. Sedimentation assays and negative-stain transmission electron microscopy revealed that efficient filament formation requires equimolar filensin–phakinin stoichiometry. Off-stoichiometric mixtures resulted in reduced filament elongation and increased non-filamentous material, particularly under phakinin-rich conditions. In addition, αB-crystallin co-sedimented with assembled filensin–phakinin filaments but not with either protein alone, and its addition altered filament morphology from smooth to beaded. These results suggest that the beaded appearance historically associated with lens-specific filaments may arise, at least in part, from association with lens crystallins. Domain truncation analysis showed that terminal regions differentially regulate filament assembly. Domain truncation analysis showed that terminal regions differentially regulate filament assembly: head-domain deletion reduced filament elongation, whereas filensin tail deletion altered filament morphology and increased filament length in vitro. Mouse lens analysis further revealed processed filensin species in both young and aged lenses, with stronger accumulation in aged lenses with reduced transparency, suggesting that filensin processing occurs in the lens and becomes more pronounced with aging. To define the core interaction module, I analyzed rod-domain complexes of filensin and phakinin. Pull-down assays showed refolding-dependent and saturable 1:1 association between the rod domains. Native PAGE, chemical cross-linking, and SEC–MALS further supported the formation of higher-order heteromeric assemblies consistent with a hetero-octameric organization. Although crystallization and cryo-EM screening did not yield high-resolution structural information, these approaches highlighted the structural flexibility and heterogeneity of the rod-domain assembly. Finally, I examined cataract-associated mutations in filensin and phakinin. Among the variants tested, phakinin E233del produced the most severe structural phenotype, shortening filaments and disrupting filament organization in B-3 cells. E233del-containing assemblies also showed reduced resistance to mechanical and thermal stress. Substitution variants E233A and E233D restored filament elongation toward the wild-type phenotype, indicating that the E233del defect is not simply caused by loss of the glutamate side chain. Rod-domain pull-down assays showed that E233del did not abolish 1:1 binding, whereas native PAGE and mass photometry revealed increased higher-order oligomer heterogeneity. These findings support a model in which E233del impairs productive filament elongation by perturbing higher-order assembly rather than by preventing initial filensin–phakinin association. Together, this thesis establishes a mechanistic framework for lens-specific intermediate filament assembly. Filensin and phakinin form an obligate heteropolymeric system whose assembly depends on balanced equimolar incorporation, rod-domain interaction, terminal-domain regulation, and association with lens protein environment. These findings provide molecular insight into how lens-specific intermediate filaments support lens structure and how their disruption may contribute to cataract-associated pathology.
