Synaptic dysfunction and neuronal loss are characteristic features of Alzheimer’s disease (AD). Recent studies observed neuronal hyperactivity in the presymptomatic AD brain and increased synaptic expression in AD neurons. However, the mechanisms of increased neuronal activity in AD and its pathological function are not well understood yet. Here, I examined neuronal activity in the early neuronal development stage using human-induced pluripotent stem cells (hiPSCs) with the familial AD (fAD) mutation (APP Swedish) and isogenic control. I observed enhanced neuronal differentiation with increased levels of mitochondrial reactive oxygen species (mtROS) in APPswe neural progenitor cells (NPCs). To verify the correlation between mtROS levels and accelerated neuronal differentiation, I divided control NPCs into two groups by ROS levels and identified whether neuronal activity is increased in the high-ROS groups. Next, I investigated the causal effect of mtROS on accelerated neuronal differentiation by treating APPswe NPCs with antioxidant EUK8. Although it was not a significant level, neuronal differentiation was alleviated in the EUK8-treated APPswe neurons. I further measured amyloid-beta (Aβ) levels as neuronal activity has been positively linked with Aβ production and it can show pathological aspects of accelerated neuronal differentiation. High-ROS neural cells secreted more Aβ compared to controls, which can lead to amyloidosis. Also, I identified similar patterns about ROS and neuronal differentiation in sporadic AD mutation, which may indicate that the role of mtROS on accelerated neuronal differentiation could be a common feature of AD-linked mutations. Taken together, my study suggests the potential contribution of accelerated neuronal differentiation by abnormal mtROS levels to neuronal hyperactivity in AD.; 알츠하이머병에서 신경세포의 사멸이 특징적으로 알려져 있지만, 병의 아주 초기 부분에 있어서는 신경세포의 과활성화 현상이 나타나는 것이 보고되었다. 본 논문은 미토콘드리아 활성산소가 신경세포의 과활성화에 영향을 주고, 알츠하이머병의 발병에 영향을 줄 수 있는지에 대해 탐구하였다. 이런 신경세포의 과활성화가 나타나는 메커니즘을 연구하기 위해 알츠하이머 가족성 유전자를 가진 인간 유래 역분화줄기세포를 이용해 발생 단계의 시점의 변화를 확인하였다. 정상인 대비 알츠하이머 유전자를 가진 신경전구세포에서 더 빠른 신경세포로의 분화가 일어나는 것과 활성산소가 높아져 있는 것을 발견하였다. 그리고 이 둘의 관계를 보기 위해 정상 세포를 활성산소 수준에 따라 낮은 그룹과 높은 그룹으로 분리하여 그 둘의 분화 과정을 비교하였다. 또한 활성산소를 줄여주는 항산화제를 처리하여 알츠하이머 신경세포에서의 가속화된 신경세포의 분화 현상이 감소하는지를 확인하였다. 그리고 알츠하이머병의 가장 주된 원인요소로 지목되고, 신경세포의 활성과 연관이 많은 아밀로이드베타가 신경세포로의 분화와 증가된 활성산소의 수준에 의해 어떻게 변화하는지를 탐구하였다. 그리고 이런 현상이 늦은 시기에 발병하는 산발성 알츠하이머에서도 나타는지를 확인해 보기 위해 반복 실험으로 양상을 비교해보았다. 그 결과, 산발성 알츠하이머에서도 더 증가된 신경 분화와 높아져 있는 활성 산소를 확인할 수 있었다. 결과를 종합해 보았을 때 미토콘드리아 활성산소가 신경세포의 가속화된 분화를 이끌 수 있다는 것을 확인하였고, 증가한 아밀로이드베타에 의한 뉴런의 활성도 증가 등을 통해 알츠하이머 초기에 관찰되는 신경세포의 과활성화에 기여할 가능성을 시사하였다. 이를 통해 미토콘드리아의 활성산소 수준의 적절한 조절이 알츠하이머 치료의 표적이 될 수 있음을 제안한다.
