- Abstract
- 본 발명은 시계 유전자 Period의 발현을 억제용 단일 가이드 RNA 조합, 및 이의 용도에 관한 것으로, 시계 유전자(clock gene)인 Period 1(Per1) 및 Period 2(Per2)의 발현을 억제할 수 있는 특정 sgRNA 조합을 설계하고, 이를 포함하는 벡터를 이용하여 시교차상핵(suprachiasmatic nucleus, SCN)에서 일주기 분자 시계 진동이 억제되는 것을 확인하고, 상기 벡터를 포함하는 바이러스를 주입한 동물 모델에서 일주일 운동 활성에 운동 활동이 낮아지고 부정맥이 나타나는 등의 일주기 리듬 억제에 의한 병증이 나타나는 것을 확인함으로써, 상기 sgRNA 조합 및 이를 포함하는 벡터를 일주기 생체 리듬을 변화시킬 수 있는 도구로 제공된다.
