dc.description.abstract Ion channels play essential roles in somatosensory processing, yet how individual channels contribute to distinct components of somatosensory circuits remains incompletely understood. The two-pore domain potassium channel Twik-1 has been implicated in sensory signaling, but its functional roles across somatosensory circuits remain unclear. Here, I demonstrated the roles of Twik-1 in spinal mechanosensory circuits and peripheral sensory neurons using constitutive and compartment-specific genetic manipulations. Constitutive deletion of Twik-1 reduced behavioral responses to innocuous tactile stimuli and von Frey-evoked punctate mechanical stimulation while leaving behavioral responses to high-intensity mechanical, heat, and cold stimuli largely unchanged. Selective deletion of Twik-1 in spinal neurons or inhibitory interneurons recapitulated these mechanosensory deficits, indicating that Twik-1 in spinal inhibitory circuits contributes to normal tactile and punctate mechanical responses. In contrast, selective deletion of Twik-1 in dorsal root ganglion neurons preserved baseline somatosensory function but impaired the persistence of nerve injury- induced mechanical hypersensitivity. This phenotype was associated with reduced aberrant excitability of injured DRG neurons and altered transcriptional responses following nerve injury. Together, these findings reveal distinct roles of Twik-1 in somatosensory circuits, supporting normal mechanosensory processing through spinal inhibitory circuits while contributing to the persistence of neuropathic pain via its functions in primary sensory neurons. These findings support further investigation of Twik-1 as a potential therapeutic target for neuropathic pain.|체성감각 시스템은 외부 환경과 신체 내부에서 발생하는 촉각, 기계적 자극, 온도 자극, 고유수용성 감각 및 유해 자극을 감지하고 이를 적절한 행동 반응으로 전환하는 신경회로이다. 이러한 감각 정보는 후근신경절에 위치한 일차 감각 뉴런에서 시작되어 척수후각의 흥분성 및 억제성 인터뉴런 회로를 거쳐 상위 중추로 전달된다. 정상적인 기계감각 처리는 말초 감각 뉴런과 척수 회로 사이의 정교한 균형에 의해 조절되며, 말초신경 손상 후에는 이러한 균형이 무너져 지속적인 기계적 과민 반응과 신경병성 통증이 발생할 수 있다.
이온 채널은 감각 뉴런과 척수 뉴런의 흥분성을 조절하는 핵심 인자이다. 그러나 하나의 이온 채널이 체성감각 회로의 서로 다른 구획에서 동일한 기능을 수행하는지, 또는 생리적 감각 처리와 병리적 통증 상태에서 서로 다른 역할을 하는지는 충분히 밝혀지지 않았다. Twik-1(Kcnk1/K2P1.1)은 누출성 칼륨 채널로, 다양한 신경세포와 교세포에서 막전위 및 세포 흥분성 조절에 관여하는 것으로 알려져 있다. 기존 연구들은 Twik-1이 후근신경절 감각 뉴런에서 신경병성 통증과 관련될 수 있음을 제시하였으나, Twik-1이 척수후각 회로와 말초 감각 뉴런에서 각각 어떤 기능을 수행하는지는 명확하지 않았다.
본 학위논문에서는 전신적, 조직 특이적, 세포유형 특이적 Twik-1 결손 마우스를 이용하여 Twik-1의 체성감각 및 신경병성 통증 조절 기능을 분석하였다. RNAscope in situ hybridization, Twik-1EGFP BAC reporter mouse, 공개 단일세포 RNA sequencing 자료의 재분석을 통해 Twik-1이 후근신경절에서는 주로 중간 및 큰 지름 감각 뉴런, 특히 저역치 기계수용체 및 고유수용성 감각 계열에 풍부하게 발현되며, 척수후각에서는 흥분성 뉴런, 억제성 뉴런, 성상교세포를 포함한 여러 세포 유형에 분포함을 확인하였다.
전신적 Twik-1 결손 마우스는 무해한 촉각 자극, 본 프레이 필라멘트에 의한 점상 기계 자극, 접착 테이프 제거 및 질감 인식에서 감소된 행동 반응을 보였으나, 고강도 기계 자극, 열 자극, 냉 자극 및 운동·불안 관련 행동은 대체로 정상적으로 유지되었다. 조직 특이적 분석에서는 DRG 뉴런의 Twik-1 결손이 기저 기계감각 결손을 유발하지 않았고, 반대로 척수 뉴런 또는 Vgat 양성 억제성 인터뉴런에서 Twik-1을 제거하거나 knockdown했을 때 전신 결손 마우스와 유사한 기계감각 저하가 나타났다. 또한 척수 억제성 인터뉴런의 전기생리학적 분석은 Twik-1 결손이 강한 입력 조건에서 반복 발화 빈도를 증가시킴을 보여주었다. 이는 Twik-1이 척수 억제성 회로의 흥분성을 제한함으로써 정상적인 촉각 및 점상 기계 자극 처리에 기여함을 시사한다.
반면, 말초신경 손상 후 신경병성 통증 상태에서는 후근신경절 뉴런의 Twik-1이 핵심적인 역할을 하였다. 만성 압박 손상 모델에서 전신적 Twik-1 결손 마우스와 후근신경절 특이적 Twik-1 결손 마우스는 초기 기계적 과민 반응을 보였으나, 시간이 지남에 따라 기계적 과민 반응의 지속성이 감소하였다. 이에 비해 척수 또는 척수 억제성 뉴런에서 Twik-1을 제거한 경우에는 신경손상 후 기계적 과민 반응이 유지되었다. 손상 후 후근신경절 뉴런의 패치 클램프과 생체 내 칼슘 이미징은 Twik-1 결손이 손상 후근신경절 뉴런의 비정상적 기계 반응성과 과흥분성을 감소시킴을 보여주었다. 또한 bulk RNA sequencing 분석은 Twik-1 결손 후근신경절에서 신경손상에 의해 유도되는 염증 반응, Integrin 과 Wnt 신호전달 및 통증 관련 전사 조절 프로그램이 변화함을 나타냈다. 추가적으로 HEK293T 세포에서의 전기생리학적 분석은 Twik-1의 이온 선택성과 전류 특성이 외부 이온 조성 및 PI(4,5)P2 의존성 신호 상태에 따라 변화할 수 있음을 보여주었다. 이는 Twik-1이 단순한 정적 누출 칼륨 채널이 아니라, 병리적 환경에서 감각 뉴런 흥분성을 상태 의존적으로 조절할 가능성을 제시한다.
종합하면, 본 연구는 Twik-1이 체성감각 회로에서 조직 특이적이고 상태 의존적인 기능을 수행함을 보여준다. 척수 억제성 회로에서 Twik-1은 정상적인 촉각 및 점상 기계 자극 처리에 기여하는 반면, 후근신경절 감각 뉴런에서 Twik-1은 말초신경 손상 후 지속적인 기계적 과민 반응과 신경병성 통증 유지에 관여한다. 이러한 발견은 하나의 이온 채널이 말초 및 중추 체성감각 회로에서 서로 다른 기능을 가질 수 있음을 보여주며, Twik-1이 신경병성 통증 조절을 위한 잠재적 치료 표적이 될 수 있음을 시사한다.
