Abstract

Neurons are highly specialized post-mitotic cells with extended axonal structures, rendering them particularly vulnerable to injury and pathological insults. To maintain functional integrity, neurons dynamically regulate gene expression across multiple molecular levels to determine survival, degeneration, regeneration, and proper development. However, it remains unclear how neurons coordinate intrinsic signaling and extrinsic cellular responses through distinct layers of gene regulation to adapt to injury and disease. This dissertation investigates gene expression regulation in neurons under both peripheral nerve injury and disease-associated pathological conditions.
In the context of peripheral nerve injury, we demonstrate that dual leucine zipper kinase (DLK), a master regulator of intrinsic neuronal stress signaling, is translationally upregulated through upstream open reading frame (uORF)-mediated control via the integrated stress response. In parallel, dipeptidyl peptidase 4 (DPP4), predominantly expressed in Schwann cells, is rapidly downregulated at the injury site while transiently increased in circulation, suggesting shedding-mediated systemic signaling. Mechanistically, our data suggest that DLK signaling may contribute to injury-induced regulation of DPP4 shedding, thereby linking intrinsic neuronal stress responses to extracellular microenvironmental remodeling during nerve regeneration. These findings support a neurocentric model in which injured neurons actively coordinate regenerative niche remodeling, rather than functioning as passive targets of glial support.
Under pathological conditions, we investigated the role of potassium channel tetramerization domain containing 3 (KCTD3), a gene associated with neurodevelopmental disorders. Loss of KCTD3 disrupted axon initial segments (AIS) and axon growth, accompanied by reduced levels of dishevelled-associated activator of morphogenesis 1 (DAAM1), a cytoskeletal regulator. Restoration of DAAM1 partially rescued the observed defects, indicating that KCTD3 regulates neuronal development through cytoskeletal modulation.
Together, these findings demonstrate that neurons employ distinct yet coordinated gene regulatory mechanisms to adapt to injury and pathological conditions. This work highlights translational control, protein shedding, and cytoskeletal regulation as key layers of neuronal gene expression regulation and provides insight into molecular pathways that may serve as potential therapeutic targets for neurodegenerative, traumatic, and neurodevelopmental disorders.|신경세포는 긴 축삭 구조를 가지는 고도로 특화된 분열불능 세포로서 외부 자극과 병리적 스트레스에 취약하다. 이러한 조건에서 신경계의 기능적 항상성을 유지하기 위해 신경세포는 다양한 수준에서 유전자 발현을 정교하게 조절하여 생존, 퇴행, 재생 반응을 결정한다. 그러나 축삭 손상 조건과 병리적 상태에서 신경세포가 어떠한 유전자 조절 기전을 통해 반응하며, 세포 내 신호와 주변 세포 간 반응이 어떻게 통합되는지는 충분히 규명되지 않았다. 본 학위논문에서는 말초신경 손상과 질환 관련 병리적 조건에서 신경세포의 유전자 발현 조절 기전을 규명하고자 하였다.
말초신경 손상 모델에서 dual leucine zipper kinase (DLK)가 손상 유도성 세포 내 스트레스 반응의 핵심 조절 인자로 작용함을 확인하였으며, DLK 발현이 단백질 분해 억제가 아닌 upstream open reading frame (uORF)에 의한 번역 수준 조절과 통합 스트레스 반응을 통해 증가함을 밝혔다. 동시에, 슈반세포에서 발현되는 dipeptidyl peptidase 4 (DPP4)는 손상 부위에서 빠르게 감소하는 반면 혈중에서는 일시적으로 증가하여 shedding을 통한 전신 반응 가능성을 시사하였다. 또한 손상 유도성 DLK 신호와 DPP4 변화 사이의 연관성을 확인함으로써, 신경세포의 내재적 스트레스 반응이 세포외 미세환경 재구성과 연계되어 재생 반응을 조절할 가능성을 제시하였다. 이러한 결과는 손상된 신경세포가 단순히 슈반세포의 도움을 받는 수동적 대상이 아니라, 재생 미세환경 형성을 능동적으로 조율하는 주체로 작용할 수 있음을 시사한다.
병리적 조건에서는 신경발달 이상과 관련된 potassium channel tetramerization domain containing 3 (KCTD3)의 기능을 분석하였다. KCTD3 결손은 축삭 성장 및 axon initial segment (AIS)의 정상적인 형성을 저해하였으며, 세포골격 조절 단백질인 dishevelled-associated activator of morphogenesis 1 (DAAM1)의 감소를 유도하였다. DAAM1 발현 회복은 이러한 표현형을 부분적으로 구제함으로써 KCTD3가 세포골격 조절을 통해 신경발달을 조절함을 시사하였다.
종합적으로 본 연구는 신경세포가 손상 및 병리적 조건에 대응하기 위해 번역 조절, 단백질 shedding, 세포골격 재구성 등 다양한 수준의 유전자 발현 조절 기전을 활용함을 제시한다. 이러한 결과는 신경 손상, 신경퇴행성 질환 및 신경발달 이상 질환의 병태생리를 이해하는 데 기초적 근거를 제공하며, 향후 치료 전략 개발을 위한 새로운 분자 표적을 제시할 것으로 기대된다.
